OSS_128167 (SIRT6-IN-1) is a specific SIRT6 inhibitor with IC50 values of 89, 1578 and 751 μM for SIRT6, SIRT1 and SIRT2, respectively. This compound has an anti-viral effect in HBV transcription and replication.

In vitro
OSS-128167 is a selective compound towards SIRT6 with IC50 values towards SIRT1 and SIRT2 being approximately 20 times higher. It is cell permeable and active in cultured cells. This compound increases H3K9 acetylation and GLUT-1 expression.
4 × 105 BxPC3 cells are plated in six-well plates and allowed to adhere for 24 h. Thereafter, cells are stimulated with 100 μM OSS-128167 or the respective amounts of vehicle DMSO for the indicated time amounts. Finally, cells are used for protein lysate generation, and total H3 and acetylated H3K9 levels are detected by immunoblotting.
